Pump-probe multi-mode super-resolution microscopy

Abstract :
 We have developed multimode super-resolution microscopy to study various bio and medical samples. The method is based on pump-probe strategy which has been extensively studied by our group for many years.  The pump-probe multi-mode imaging is composed of two types of modes; stimulated emission (stimulated emission imaging microscope, SEIM) mode and photothermal (photothermal microscope, PTM) mode. The former has two sub modes. They are stimulated emission induced gain (SEIG) and stimulated emission induced quenching (SEIQ).

 Using then we could obtain 140 nm resolution and in principle by the method we can reach 70 nm by combination of other principles. We have taken images of such as mouse brain, rat muscle, mouse kidney and, mouse skin cancer.

The advantages of our method are (1) super-resolution, (2) compact, (3) applicable to nonfluorescent sample, (4) high sensitivity, (5) capability of multi-color imaging.
